Can I keep a managed Aperture Library file on an external hard drive (Drobo S) which will be always connected to my iMac and will the Drobo be fast enough. Or will it have to be referenced by the fact of it not being internal

You can store your Aperture library, managed or referenced, wherever you like. So yes, you can have a managed library on your Drobo. If it’s connected by FW800 (or eSATA on a Mac Pro), it’ll be fast enough. USB2 is not fast enough… I mean, technically it’ll work, but that doesn’t make it a Good Idea™.
Good luck, and you may want to consider picking up the “In-Depth Getting Started with Aperture 3” eBook if you haven’t already. It’ll help explain many of the basics of Aperture, giving you a really good foundation for future growth.

I think I will have a go at managed then as long as I have Drobo plugged in Aperture looks after the rest all in one place.
